基于核酸适配体免标记荧光法检测卡那霉素的研究

Label-free kanamycin detection based on nucleic acid aptamers

  • 摘要: 基于卡那霉素核酸适配体(Apt-DNA)和SYBR Green I(SG)建立了一种免标记荧光检测卡那霉素的方法。Apt-DNA与其部分互补的寡核苷酸链(Pri-DNA)通过碱基配对形成双链DNA(dsDNA)。游离态的SG发出微弱的荧光, 嵌入dsDNA后SG的荧光大幅度增强。溶液中存在卡那霉素时, Apt-DNA与卡那霉素特异性结合, dsDNA浓度减少, 荧光强度也随之下降。通过对比卡那霉素存在前后的荧光强度的变化, 实现了卡那霉素的免标记检测。该方法具有设计原理简单, 操作便捷, 灵敏度高, 成本较低等优点, 并且成功应用于实际样品中卡那霉素的检测。

     

    Abstract: A fluorescent and label-free assay for the detection of kanamycin was developed based on kanamycin aptamer (Apt-DNA) and SYBR Green I (SG). Apt-DNA combines with complementary oligonucleotide chain (Pri-DNA) to form double stranded DNA (dsDNA). Although the free-state SG emits a weak fluorescence, the fluorescence intensity is significantly enhanced after attaching to dsDNA. When kanamycin was added into the solution, Apt-DNA specifically bind with kanamycin and the concentration of dsDNA is dramatically reduced, which obviously decrease the fluorescence intensity. By comparing the fluorescence intensity before and after adding kanamycin, the label-free detection of kanamycin is realized. Some advantages of this method are presented, such as simple principle, convenient operation, high sensitivity and low cost. What' more, the proposed method could be used to measure kanamycin in real samples.
