新型纳米磁色谱微流体芯片
A novel magnetic chromatographic microsystem

摘要: 通过标准光刻电铸工艺,在玻璃基片上制备了不同形状的顺磁性镍铁微柱阵列作为微流控通道内的磁力元件;通过SU-8胶光刻模板PDMS快速成型制备了微流控通道结构,通过O2 Plasma处理和显微镜下对准,实现在玻璃基片上的永久封装,制备了磁分离微流控芯片;在微流控通道中引入含有微磁珠的溶液,通过外加磁场和流动式进样,观察微磁珠的对磁场的响应情况以及层流中的磁珠分离情况及捕获,并进行了DNA提取实验。Abstract: beads used as solid supports for biomedical in microfluidics have gained more attraction in recent years.However,controling and manipulating magnetic beads in microfluidics is still a problem.Here we demonstrate a novel method for immobolizing the magnetic beads and using the beads as solid phase for chromotography in microfluidics.NiFe micropillar array higher than 20um was fabricated inside the microchannel in order to generate strong magnetic field gradients and capture the magnetic beads even at a high flow rates.Captured magnetic beads would serve as a solid phase for chromatography.DNA purification were tested by this chip.The magnetic beads based chromatography method would also be used in enzyme reaction,protein purification and provide a new way for manipulate the bimolecular in microfluidics.
